In Vivo 功能級抗體是專為活體動物研究所設計的高純度抗體。與一般研究用抗體不同,功能級抗體不含防腐劑、穩(wěn)定劑或其他添加物,并針對活體內(nèi)的生物活性與安全性進行優(yōu)化。這類抗體常用于臨床前研究中,以達到免疫細胞去除、阻斷受體與配體間的交互作用,或啟動特定信號傳導途徑的目的。其來源通常為雜交瘤細胞培養(yǎng)或重組表達系統(tǒng),并經(jīng)過完善效能驗證。
In Vivo 用途抗體的重要特性
為確保實驗再現(xiàn)性、降低副作用風險,并能在動物模型中進行全身性注射而不引發(fā)非特異性免疫反應,In Vivo 功能級抗體必須符合嚴格的品質(zhì)標準:
低內(nèi)毒素:內(nèi)毒素含量需嚴格控制,通常低于 2 EU/mg,以避免引發(fā)炎癥或免疫活化。
無載體蛋白:不添加 BSA、明膠或其他蛋白質(zhì),以降低非預期免疫反應的風險。
無防腐劑:不含 Sodium Azide 或 Thimerosal,以確保在體內(nèi)使用的安全性。
特定抗體 IgG 子類別:可依需求選擇合適的 IgG 類型,以發(fā)揮所需的 Fc 介導功能。
高穩(wěn)定性與高純度:通過親和純化或 Protein A/G 層析純化,并經(jīng) SDS-PAGE 與功能性測試確認品質(zhì)。
動物模型中的常見應用
1. 免疫細胞去除(Immune Cell Depletion)
針對特定免疫細胞標記的抗體可用于短暫去除目標細胞群,以評估其在疾病或生理平衡中的作用。
