脂質(zhì)立方相(LCP)結(jié)晶已經(jīng)發(fā)展成為膜蛋白結(jié)晶的一種重要方法,其中脂質(zhì)單油酸甘油酯(Monoolein)是創(chuàng)建穩(wěn)定LCP的首選。然而,在過去幾年中,短鏈脂質(zhì)7.7 MAG[4,7]、7.8 MAG[6-8]和7.9 MAG[5,7]變得越來越受歡迎。最近在將高黏度脂質(zhì)立方相應(yīng)用于連續(xù)飛秒晶體學(xué)(LCP-SFX)方面的進(jìn)展[1-3],通過以下方式進(jìn)一步加速了LCP方法的成功:


  • 將載有晶體的黏性LCP直接送入X射線自由電子激光(XFEL)光束中(與液體注射器相比,減少了樣品消耗);

  • 利用在LCP中生長(zhǎng)的固有小晶體;

  • 避免了從LCP中進(jìn)行棘手的晶體裝載。

JBScreen LCP旨在高效篩選LCP中的結(jié)晶條件。上述表格中的每種LCP脂質(zhì)都能創(chuàng)建一個(gè)穩(wěn)定的脂質(zhì)立方相,可用于與JBScreen LCP進(jìn)行結(jié)晶設(shè)置。其96種條件是通過對(duì)192種成功通過in meso方法結(jié)晶并產(chǎn)生結(jié)構(gòu)的整合膜蛋白進(jìn)行數(shù)據(jù)挖掘得出的[7]。該篩選按沉淀劑的濃度和類型排序,并且不含乙酸鎘。
