W3/25單克隆抗體與大鼠CD4發(fā)生反應。CD4是免疫球蛋白超家族的55 kDa I型膜糖蛋白，作為細胞表面受體蛋白，它充當抗原呈遞細胞（APC）顯示的II類MHC分子的共受體（與TCR）。CD4由大多數(shù)胸腺細胞、輔助T細胞、NK-T細胞亞群表達，由樹突狀細胞和巨噬細胞弱表達。CD4在T細胞的發(fā)育中起著重要作用，是成熟T細胞最佳功能所必需的。克隆W3/25被用作“輔助性T細胞標記物”。克隆W3/25是一種不完全的單克隆抗體，但已被證明在體外中和CD4。幾項體外研究還表明，W3/25單克隆抗體通過下調(diào)淋巴細胞表面的CD4分子來抑制CD4+T細胞活化。W3/25單抗還在MLR實驗中抑制抗原誘導的T細胞增殖和IL2的產(chǎn)生。在預防性和實驗性治療性體內(nèi)研究中，該抗體已被廣泛用于調(diào)節(jié)實驗性變態(tài)反應性腦脊髓炎（EAE）、佐劑性關節(jié)炎等疾病相關和機制參數(shù)。
