檢測原理：

雙抗體夾心ELISA的原理如圖1所示。在該測定中，樣品中存在的IgA與已吸附在聚苯乙烯微量滴定池表面的抗IgA抗體反應(yīng)。在通過洗滌去除未結(jié)合的蛋白質(zhì)后，加入與辣根過氧化物酶（HRP）綴合的抗IgA抗體。這些酶標記的抗體與先前結(jié)合的IgA形成復(fù)合物。在另一個洗滌步驟之后，通過添加無色底物3，3’，5，5’-四甲基聯(lián)苯胺（TMB）來測定與免疫吸附結(jié)合的酶。結(jié)合酶的數(shù)量與所測試樣品中IgA的濃度直接相關(guān)；因此，在450nm處的吸光度是測試樣品中IgA濃度的測量值。試驗樣品中IgA的量可以根據(jù)標準曲線進行插值，并根據(jù)樣品稀釋度進行校正。
