Botin修飾的脫氧腺苷5'-三磷酸被廣泛用于各種非放射性DNA標(biāo)記反應(yīng),包括缺口翻譯、隨機(jī)引物標(biāo)記、cDNA標(biāo)記和3'-末端標(biāo)記。生物素化探針已被證明與同源核酸雜交的速度和程度與非生物素化的探針相同。雜交的生物素化DNA探針可以用抗生物素蛋白和鏈霉抗生物素素蛋白檢測(cè)。Tris緩沖液(pH 7.5)中的生物素-11-dATP*1 mM*可以通過(guò)缺口翻譯、隨機(jī)引物、3'-末端標(biāo)記或PCR過(guò)程酶促摻入DNA。數(shù)字“11”是指dATP和生物素之間連接體骨架中的碳原子數(shù)。生物素和抗生物素蛋白之間更有效的相互作用將發(fā)生在較長(zhǎng)的連接體上,而較短的連接體將導(dǎo)致dATP更有效地?fù)饺隓NA。建議接頭'11'長(zhǎng)度對(duì)于大多數(shù)應(yīng)用來(lái)說(shuō)是最佳的。Tris緩沖液(pH 7.5)中的生物素-11-dATP*1 mM*在各種雜交應(yīng)用中產(chǎn)生生物素化的DNA探針,包括Southern印跡、Northern印跡、斑點(diǎn)印跡、固定細(xì)胞和組織。其化學(xué)性質(zhì)與PerkinElmer(PE)的NEL540001EA相當(dāng)。
