C反應(yīng)蛋白(CRP)是肝臟對炎癥反應(yīng)產(chǎn)生的一種物質(zhì)。它是身體炎癥的標(biāo)志物,經(jīng)常在醫(yī)療環(huán)境中用于評估炎癥的存在和嚴(yán)重程度。CRP水平會因各種情況而升高,包括感染、損傷和慢性炎癥性疾病。
血液中高水平的CRP可以表明炎癥的存在,但不能確定確切的原因。它通常與其他臨床和實驗室發(fā)現(xiàn)結(jié)合使用,以幫助診斷和監(jiān)測感染、自身免疫性疾病和心血管疾病等疾病。CRP水平升高也可能與類風(fēng)濕性關(guān)節(jié)炎、炎癥性腸病和某些癌癥等疾病有關(guān)。
在雙抗體夾心狗/犬CRP ELISA測定中,樣品中存在的CRP與已吸附到聚苯乙烯微孔表面的抗狗CRP抗體反應(yīng)。在通過洗滌去除未結(jié)合的蛋白質(zhì)后,加入與辣根過氧化物酶(HRP)綴合的抗狗CRP抗體。這些酶標(biāo)記的抗體與先前結(jié)合的CRP形成復(fù)合物。在另一個洗滌步驟之后,通過添加顯色底物3,3’,5,5’-四甲基聯(lián)苯胺(TMB)來測定與免疫吸附結(jié)合的酶。結(jié)合酶的量直接隨測試樣品中犬CRP的濃度而變化;因此,在450nm處的吸光度是測試樣品中CRP濃度的測量值。試驗樣品中CRP的量可以根據(jù)標(biāo)準(zhǔn)曲線進(jìn)行插值,并根據(jù)樣品稀釋度進(jìn)行校正
