IRE1-XBP1細(xì)胞應(yīng)激測定#U0921G：

使用明亮的IRE1-XBP1比率電池應(yīng)力傳感器在讀板器上檢測應(yīng)力響應(yīng)。CMV啟動(dòng)子。

該傳感器基于在內(nèi)質(zhì)網(wǎng)應(yīng)激期間由IRE1蛋白剪接的XBP1 mRNA。當(dāng)細(xì)胞經(jīng)受內(nèi)質(zhì)網(wǎng)（ER介導(dǎo)的）應(yīng)激或經(jīng)歷未折疊蛋白反應(yīng)（UPR）時(shí)，紅色熒光蛋白組成性表達(dá)，而傳感器產(chǎn)生非常亮的綠色熒光。只使用綠色熒光測量應(yīng)力，或者取綠色和紅色通道的比例。此外，紅色和綠色熒光蛋白都是核定位的，用于簡單的細(xì)胞分割和圖像分析。
