想象一下,當(dāng)你回到家坐在沙發(fā)上, 看到茶幾上有一塊鮮紅多汁的西瓜,于是你拿起西瓜大塊朵頤。事實上,這個簡單的場景包含了無數(shù)神經(jīng)突觸的協(xié)助。
我們都知道,神經(jīng)信息的傳遞是靠一個神經(jīng)元傳遞給下一個神經(jīng)元的方式完成的。當(dāng)視覺神經(jīng)元接收到西瓜的信號后,神經(jīng)元的軸突末梢,也就是突觸小體,會與多個神經(jīng)元的細胞體或樹突相接觸,形成突觸,這種突觸是由突觸前膜、突觸間隙和突觸后膜三部分構(gòu)成。突觸前神經(jīng)元的軸突頂枝釋放突觸囊泡,其內(nèi)含物就是神經(jīng)遞質(zhì),神經(jīng)遞質(zhì)將信號傳遞給下一個神經(jīng)元的細胞體或樹突,完成信號的單向傳遞;最后到達大腦,接收到神經(jīng)元傳遞過來的信號,大腦做出指示,然后再一次通過神經(jīng)突觸的協(xié)助,完成后續(xù)動作。

突觸囊泡循環(huán)是突觸事件中最核心的部分,突觸囊泡通過胞吐和內(nèi)吞作用完成一次突觸囊泡循環(huán)。當(dāng)突觸囊泡膜與突觸前膜融合時, 遞質(zhì)釋放, 即為胞吐; 囊泡膜與質(zhì)膜分離重新內(nèi)陷,稱為胞吞。突觸囊泡循環(huán)主要包括以下九個步驟:錨靠、激活、融合/出胞、入胞、移位、內(nèi)質(zhì)體融合、出芽、神經(jīng)遞質(zhì)攝取、囊泡移回突觸前膜的活化區(qū)。

與經(jīng)典神經(jīng)-肌肉接頭突觸和中樞神經(jīng)元常型突觸相比較,還存在一種特殊的神經(jīng)突觸——緞帶突觸或稱帶狀突觸(ribbon synapse)。帶狀突觸被發(fā)現(xiàn)存在于視網(wǎng)膜的感光細胞和雙極細胞,以及內(nèi)耳前庭毛細胞和耳蝸內(nèi)毛細胞。視網(wǎng)膜及內(nèi)耳的帶狀突觸通過緊張性釋放神經(jīng)遞質(zhì)傳導(dǎo)不同強度的光和聲音信息。
與中樞神經(jīng)系統(tǒng)突觸的胞吐機制相似,帶狀突觸囊泡釋放也分為兩種:快速釋放池(readily releasable pool,RRP)和慢速釋放池(slowly releasable pool,SRP),但是帶狀突觸遞質(zhì)釋放較經(jīng)典海馬神經(jīng)元突觸囊泡釋放速率明顯增快。
