- The Complement Factor B (CFB) ELISA Kit (Monkey) is a 1.5-hour solid-phase competitive ELISA designed for the quantitative determination of CFB in monkey serum, plasma, cell culture supernatants, tissue homogenates and body fluids. This kit is intended for research use only and may not be used for therapeutic or diagnostic applications.Detection Range:2.5-50ng/mlSensitivity:0.1ng/mlPrecision:Intra-Assay CV: 5.7, 7.5%Inter-Assay CV: 4.2, 5.6%Specificity:This assay has high sensitivity and excellent specificity for detection of CFB. No significant crossreactivity or interference with analogs was observed.?Note: Crossreactivity detection between CFB and all analogs has not been determined and crossreaction may exist in some cases.Assay Principle:Complement Factor B (CFB) BioAssay ELISA Kit (Monkey) utilizes the competitive immunoassay technique utilizing a monoclonal anti-CFB antibody and a CFB-HRP conjugate. The assay sample and buffer are incubated together with CFB-HRP conjugate in the pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to halt the reaction, which will then turn the solution yellow. The intensity of the color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CFB concentration since CFB from samples and CFB-HRP conjugate compete for the anti-CFB antibody binding site. Since the number of sites is limited, as more sites are occupied by CFB from the sample, fewer sites are left to bind CFB-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CFB concentration in each sample is interpolated from the standard curve.Kit Components:189282A: Microtiter Strips, 1x48 wells or 1x96 wells. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
