- Cytochrome c Oxidase subunit 7C, Mitochondrial (COX7C) BioAssay ELISA Kit (Mouse) is a 1.5 hour solid-phase competitive ELISA for the quantitative determination of COX7C in mouse serum, plasma, cell culture supernatants, body fluid and tissue homogenates.Detection Range:0.5-10ng/mlSensitivity:0.1ng/mlPrecision:Intra-Assay CV: <10%Inter-Assay CV: <10%Specificity: This assay has high sensitivity and excellent specificity for detection of COX7C (Mouse). For maximum recovery of product, centrifuge the original vial prior to removing the cap.Assay Principle:Cytochrome c Oxidase subunit 7C, Mitochondrial (COX7C) BioAssay ELISA Kit (Mouse) utilizes the quantitative Competitive Enzyme Immunoassay technique,?utilizing a monoclonal antibody and an conjugate to COX7C (Mouse). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the COX7C concentration since?COX7C?from samples and COX7C-HRP conjugate compete for the anti-COX7C antibody binding site. Since the number of sites is limited, as more sites are occupied by?COX7C?from the sample, fewer sites are left to bind the COX7C-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards.
