- The BioAssay?ELISA Kit is a 1.5 hour solid-phase Competitive ELISA designed for the quantitative determination of Malondialdehyde (ICAM-1) in rabbit serum, plasma, cell culture supernatants, body fluid and tissue homogenates.Detection Range:2.5-50ng/mlSensitivity:0.1ng/mlPrecision:Intra-Assay CV: <10%Inter-Assay CV: <10%Assay Principle:Intercellular Adhesion Molecule 1 (ICAM-1) BioAssay ELISA Kit (Rabbit) utilizes the quantitative Competitive Enzyme Immunoassay technique,?utilizing a monoclonal antibody and an conjugate to ICAM-1 (Rabbit). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ICAM-1 since?ICAM-1?from samples and?ICAM-1-HRP conjugate compete for the anti-Protein?antibody binding site. Since the number of sites is limited, as more sites are occupied by?Protein?from the sample, fewer sites are left to bind the?Protein-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The?ICAM-1 concentration in each sample is interpolated from this standard curve.Specificity:This assay has high sensitivity and excellent specificity for detection of Intercellular Adhesion Molecule 1 (ICAM-1) (Rabbit). No significant crossreactivity or interference with analogs was observed.
