- Interleukin 10 (IL-10) BioAssay ELISA Kit (Porcine) is a 1.5 hour solid-phase Competitive ELISA designed for the quantitative determination of Interleukin 10 (IL-10) (Porcine) in serum, plasma, cell culture supernatants, cell lysates, body fluid and tissue homogenates.Detection Range:50-1000pg/mlSensitivity:1.0pg/mlPrecision:Intra-Assay CV: <10%Inter-Assay CV: <10%Specificity:This assay has high sensitivity and excellent specificity for detection of Interleukin 10 (IL-10) (Porcine). For maximum recovery of product, centrifuge the original vial prior to removing the cap.Assay Principle:Interleukin 10 (IL-10) BioAssay ELISA Kit (Porcine) utilizes the quantitative Competitive Enzyme Immunoassay technique,?utilizing a monoclonal antibody and an conjugate to Interleukin 10 (IL-10) (Porcine). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-10 concentration since?IL-10?from samples and?IL-10-HRP conjugate compete for the anti-IL-10?antibody binding site. Since the number of sites is limited, as more sites are occupied by?IL-10?from the sample, fewer sites are left to bind the?IL-10-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The?IL-10?concentration in each sample is interpolated from this standard curve.
