- The BioAssay?ELISA Kit is a 1.5 hour solid-phase Competitive ELISA designed for the quantitative determination of Interleukin 36 (IL-36) in mouse serum, plasma, cell culture?supernatant, tissue homogenates and body fluids. This kit is intended for research use only and may not be used for therapeutic or diagnostic applications.Detection Range:50-1000pg/mlSensitivity:1pg/mlPrecision:Intra-Assay %CV: <10Inter-Assay %CV: <10Specificity:This assay has high sensitivity and excellent specificity for detection of IL-36 (Mouse). No significant crossreactivity or interference with analogs was observed. Note: Crossreactivity detection between IL-36 and all analogs has not been determined and crossreaction may exist in some cases..Assay Principle:Interleukin 36 (IL-36) BioAssay ELISA Kit (Mouse) utilizes the quantitative Competitive Enzyme Immunoassay technique,?utilizing a monoclonal antibody and an conjugate to Interleukin 36 (IL-36) (Mouse). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-36 concentration since?IL-36?from samples and IL-36-HRP conjugate compete for the anti-IL-36?antibody binding site. Since the number of sites is limited, as more sites are occupied by?IL-36?from the sample, fewer sites are left to bind the?IL-36-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The?IL-36?concentration in each sample is interpolated from this standard curve.Kit Components:195346A: Microtiter Strips, 1x48 wells or 1x96 wells.
