- The L-Lactate Dehydrogenase B Chain (LDHB) BioAssay ELISA Kit (Rat) is a 1.5 hour solid-phase Competitive Enzyme Immunoassay designed for the quantitative determination of L-Lactate Dehydrogenase B Chain (LDHB) (Rat) in serum, plasma, cell culture supernatants, body fluid and tissue homogenates.Detection Range:25-500mU/mlSensitivity:1.0mU/mlPrecision:Intra-Assay CV: <10%Inter-Assay CV: <10%Specificity:This assay has high sensitivity and excellent specificity for detection of L-Lactate Dehydrogenase B Chain (LDHB) (Rat). For maximum recovery of product, centrifuge the original vial prior to removing the cap.Assay Principle:L-Lactate Dehydrogenase B Chain (LDHB) BioAssay ELISA Kit (Rat) utilizes the quantitative Competitive Enzyme Immunoassay technique,?utilizing a monoclonal antibody and an conjugate to L-Lactate Dehydrogenase B Chain (LDHB) (Rat). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the LDHB concentration since?LDHB?from samples and?LDHB-HRP conjugate compete for the anti-LDHB?antibody binding site. Since the number of sites is limited, as more sites are occupied by?LDHB?from the sample, fewer sites are left to bind the?LDHB-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The?Target Protein concentration in each sample is interpolated from the standard curve.
