- Nuclear Factor Erythroid 2-related Factor 2 (Nrf2) BioAssay ELISA Kit is a 1.5 hour solid-phase competitive ELISA designed for the quantitative determination of Nuclear Factor Erythroid 2-related Factor 2 (Nrf2) in human serum, plasma, cell culture supernatants, tissue homogenates and body fluids. This kit for research use only, not for therapeutic or diagnostic applications.Detection Range:0.5-10ng/mlSensitivity:0.1ng/mlPrecision:Intra-Assay %CV: <10Inter-Assay %CV: <10Specificity:This assay has high sensitivity and excellent specificity for detection of Nrf2. No significant crossreactivity or interference with analogs was observed.Note: Crossreactivity detection between Nrf2 and all analogs has not been determined and crossreaction may exist in some cases.Assay Principle:Nuclear Factor Erythroid 2-related Factor 2 (Nrf2) BioAssay ELISA Kit (Human) utilizes the quantitative Competitive Enzyme Immunoassay technique,?utilizing a monoclonal antibody and a conjugate to Nuclear Factor Erythroid 2-related Factor 2 (Nrf2) (Human). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Target Protein concentration since?Protein?from samples and?Protein-HRP conjugate compete for the anti-Protein?antibody binding site. Since the number of sites is limited, as more sites are occupied by?Protein?from the sample, fewer sites are left to bind the?Protein-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards.
