- The Sex Hormone Binding Globulin (SHBG) BioAssay ELISA Kit (Rabbit) is a 1.5-hour solid-phase competitive ELISA designed for the quantitative determination of SHBG in rabbit serum, plasma and other biological fluids. This kit is intended for research use only and may not be used for therapeutic or diagnostic applications.Kit Detection Range:100-2500pg/mlKit Sensitivity:1.0pg/mlPrecision:Intra-Assay CV: <10%Inter-Assay CV: <10%Assay Principle:This ELISA kit employs the quantitative competitive enzyme-linked immunoassay technique, utilizing a microtiter plate that has been pre-coated with a monoclonal anti- SHBG antibody. The assay samples and standards are incubated together with SHBG-HRP conjugate for one hour. A competition for limited antibody binding sites on the plate occurs between SHBG-HRP conjugate and SHBG in the samples and standards. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to terminate the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SHBG concentration in the sample or standard. A standard curve is plotted relating the intensity of the color (O.D.) to the concentrations of the standards. The SHBG concentration in each sample is interpolated from this standard curve.Kit Components:203720A: Microtiter Strips, 1x48 wells or 1x96 wells. Pre-coated with anti-SHBG antibody.203720B: Enzyme Conjugate, 1x3ml for 48Tests; 1x6ml for 96Tests.203720C: Standard A, 0pg/ml, 1x1vial203720D: Standard B, 100pg/ml, 1x1vial203720E: Standard C, 250pg/ml, 1x1vial203720F: Standard D, 500pg/ml, 1x1vial203720G: Standard E, 1000pg/ml, 1x1vial203720H: Standard F, 2500pg/ml, 1x1vial203720J: Substrate A, 1x6ml203720K: Substrate B, 1x6ml203720L: Stop Solution, 1x6ml203720M: Wash Solution, 100X, 1x10ml203720N: Balance Solution, 1x3mlStorage and Stability:Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Protocol:Please read Reagent Preparation above before starting assay procedure. It is recommended that all Standards and Samples be added in duplicate to the microtiter plate. It is strongly recommended to do a preliminary experiment before measuring all samples.1. Secure the desired number of coated wells in the holder. Add 100ul of Standards or Samples to the appropriate well in the antibody-coated microtiter strip. Add 100ul of PBS (pH 7.0-7.2) in the blank control well.2. Dispense 10ul of Balance Solution into 100ul samples; mix well.Note: This step is required when the sample is body fluid; if the sample is serum or plasma, this step should be skipped.3. Add 50ul of Enzyme Conjugate to each well (do NOT add to blank control well). Mix well. Mixing well in this step is important. Cover and incubate the plate for 1 hour at 37°C.4. Wash the microtiter plate using one of the specified methods indicated (manual or automatic).a) Manual Washing: Remove incubation mixture by aspirating contents of the plate into a sink or proper waste container. Fill in each well completely with 1x wash solution, and then aspirate contents of the plate into a sink or proper waste container. Repeat this procedure five times for a total of five washes. After washing, invert plate, and blot dry by hitting the plate onto absorbent paper or paper towels until no moisture appears. Note: Hold the sides of the plate frame firmly when washing the plate to assure that all strips remain securely in frame. Complete removal of liquid at each step is essential to good performance.b) Automated Washing: Wash plate five times with diluted wash solution (350-400ul per well per wash) using an auto washer. After washing, dry the plate as in (a). It is recommended that the washer bet set for a soaking time of 10 seconds and shaking time of 5 seconds between each wash.5. Add 50ul of Substrate A and 50ul of Substrate B to each well including blank control well. Cover and incubate for 10-15 minutes at 37°C. (Avoid sunlight).6. Add 50ul of Stop Solution to each well including blank control well. Mix well.7.
