- Assay Principle:Tartrate-resistant Acid Phosphatase 5b (TRACP-5b) BioAssay ELISA Kit (Rat) utilizes the quantitative Competitive Enzyme Immunoassay technique, utilizing a monoclonal antibody and a conjugate to Tartrate-resistant Acid Phosphatase 5b (TRACP-5b) (Rat). The assay sample and buffer are incubated together with the HRP conjugate in a pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Target Protein concentration since?Protein?from samples and?Protein-HRP conjugate compete for the anti-Protein?antibody binding site. Since the number of sites is limited, as more sites are occupied by?Protein?from the sample, fewer sites are left to bind the?Protein-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The?Target Protein concentration in each sample is interpolated from this standard curve.Intended Use:The BioAssay?ELISA Kit is a 1.5 hour solid-phase competitive ELISA designed for the quantitative determination of Tartrate-resistant Acid Phosphatase 5b (TRACP-5b) in rat serum, plasma, cell culture?supernatant, tissue homogenates and body fluids. This kit is for research use only, not for therapeutic or diagnostic applications.Detection Range:2.5-50ng/mlSensitivity:0.1ng/mlPrecision:Intra-Assay %CV: 7.1, 9.4Inter-Assay %CV: 5.3, 7.9Specificity:This assay has high sensitivity and excellent specificity for detection of Tartrate-resistant Acid Phosphatase 5b (TRACP-5b). No significant crossreactivity or interference with analogs was observed.
