- This human Angiopoietin-1 BioAssay ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of human Angiopoietin-1. Sample Type:Serum, plasma, cell culture supernatants and urineIntended Use:For quantitative protein detection, establishing normal range and validation of antibody array results.Sensitivity:The minimum detectable dose of Angiopoietin is typically <30pg/ml.Range:30-18,000pg/mlSpecificity:Recognizes human Angiopoietin-1. This ELISA kit shows no cross-reactivity with any of the cytokines tested (e.g., human Angiogenin, Angiopoietin 2, BDNF, BLC, ENA-78, FGF-4, IL-1a, IL-1b, IL-2, IL-3, IL-4, IL-5, IL-8, IL-9, IL-10, IL- 11, IL-12 p70, IL-12 p40, IL-13, IL-15, IL-309, IP-10, G-CSF, GM-CSF, IFN-g, Leptin, MCP-1, MCP-2, MCP-3, MDC, MIP-1a, MIP-1b, MIP-1d, PARC, PDGF, RANTES, SCF, TARC, TGF-b, TIMP-1, TIMP-2, TNF-a, TNF-b, TPO, VEGF).Test Principle:This assay employs an antibody specific for human Angiopoietin-1 coated on a 96-well plate. Standards and samples are pipetted into the wells and Angiopoietin-1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-human Angiopoietin-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Angiopoietin-1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450nm. The intra-Assay reproducibility is CV<10% & inter-Assay is CV<12%.Kit Components:Microplate (Item A) coated with anti-human Angiopoietin-1, 1x96 wells?Wash Buffer Concentrate (20x) (Item B), 1x25ml?Standards (Item C) (recombinant human Angiopoietin-1), 2x vials?Assay Diluent A (Item D) Standard/Sample (serum/plasma) in 0.09% sodium azide, 1x30ml?Assay Diluent B (Item E) Standard/Sample (cell culture medium/urine). (5x conc.), 1x15ml?Detection Antibody Angiopoietin-1 (Item F), biotinylated anti-human Angiopoietin-1, 2 vials?HRP-Streptavidin Concentrate (Item G), 500x concentrated, 1x200ul?TMB One-Step Substrate Reagent (Item H) 3,3’,5,5’-tetramethylbenzidine in buffer, 1x12ml ?Stop Solution (Item I), 0.2 M sulfuric acid, 1x8mlStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
