- Internucleosomal DNA fragmentation is a hallmark of apoptosis in mammalian cells.Sample Type:Cell culture (adherent and suspension), parafin embedded tissue sections Intended Use:The TUNEL-based assay kit provides complete components including positive and negative control cells for convenient detection of DNA fragmentation in cultured cells and tissue sections.Specificity:Recognizes ApoBrdU. Species Crossreactivity: mammalianTest Principle:The Apo-BrdU In Situ DNA Fragmentation BioAssay Kit provides complete components including positive and negative control cells for conveniently detecting DNA fragmentation by fluorescence microscopy or flow cytometry. The kit utilizes Br-dUTP (bromolated deoxyuridine triphosphate nucleotides) which is more readily incorporated into DNA strand breaks than other larger ligands (e.g., fluorescein, biotin or digoxigenin). The greater incorporation gives rise to brighter signal when the Br-dUTP sites are identified by a fluorescein labeled anti-BrdU monoclonal antibody.Kit Components:Positive Control Cells, 1x5ml ?Negative Control Cells, 1x5ml Wash Buffer, 1x120ml ?Reaction Buffer, 1x0.6ml ?TdT Enzymes, 1x45ul ?Br-dUTP, 1x0.48ml ?Rinse Buffer, 1x120ml ?Anti-BrdU-FITC Antibody, 1x0.3ml ?PI/RNase Staining Buffer, 1x30ml Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
