- This human Cancer Antigen 125 (CA-125) BioAssay ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of human CA-125. Sample Type:Serum, plasma, cell culture supernatants and urineIntended Use:For quantitative protein detection, establishing normal range and validation of antibody array results.Sensitivity:The minimum detectable concentration of CA-125 is <0.6U/ml. Range:0.6-400U/mlSpecificity:Recognizes human Cancer Antigen 125. This ELISA kit shows no cross-reactivity with the following cytokines tested: human Angiogenin, BDNF, BLC, CNTF, ENA-78, FGF-4, IL-1a, IL-1b, IL-2, IL-3, IL-4, IL-5, IL-6, IL-7, IL-8, IL-9, IL-11, IL-12 p70, IL-12 p40, IL-13, IL-15, IL-309, IP-10, FGF-4, FGF-6, FGF-7, G-CSF, GDNF, GM-CSF, IFN-g, IGFBP-2, IGF-BP-3, IGF-BP-4, Leptin (OB), MCP-1, MCP-2, MCP-3, MDC, MIF, MIG, MIP-1a, MIP-1b, MIP-1d, PARC, PDGF, RANTES, SCF,SDF-1alpha, TARC, TGF-b, TIMP-1, TIMP-2, TNF-a, TNF-b, TPO, VEGF.Test Principle:This assay employs an antibody specific for human CA-125 coated on a 96-well plate. Standards and samples are pipetted into the wells and CA-125 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-human CA-125 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CA-125 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450nm. The intra-Assay reproducibility is CV<10% & inter-Assay is CV<12%.Kit Components:Microplate (Item A) coated with anti-human CA-125, 1x96 wells?20X Wash Buffer Concentrate (Item B), 1x25ml ?Standards (Item C) (recombinant human CA-125), 2x vials Assay Diluent (Item E) Standard/Sample (serum/plasma/cell culture/urine) (5X), 1x15ml ?Detection Antibody CA-125 (Item F), biotinylated anti-human CA-125, 2x vials ?HRP-Streptavidin Concentrate (Item G), 800x concentrated, 1x200ul ?TMB One-Step Substrate Reagent (Item H) 3,3’,5,5’-tetramethylbenzidine in buffer, 1x12ml ?Stop Solution (Item I), 0.2M sulfuric acid, 1x8ml Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
