- This human Carcinoembryonic Antigen (CEA) BioAssay ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of human CEA. Sample Type:Serum, plasma, cell culture supernatants and urineIntended Use:For use in quantitative protein detection, establishing normal range and validation of antibody array results.Sensitivity:The minimum detectable concentration of CEA is <200pg/ml. Range:0.2-250ng/mlSpecificity:Recognizes human Carcinoembryonic Antigen. This ELISA kit shows no cross-reactivity with the following cytokines tested: human Angiogenin, BDNF, BLC, ENA-78, FGF-4, IL-1a, IL-1b, IL-2, IL-3, IL-4, IL-5, IL-6, IL-7, IL-8, IL-9, IL-11, IL-12 p70, IL-12 p40, IL-13, IL-15, IL-309, IP-10, G-CSF, GM-CSF, IFN-g, Leptin (OB), MCP-1, MCP-2, MCP-3, MDC, MIP-1a, MIP-1b, MIP-1d, PARC, PDGF, RANTES, SCF, TARC, TGF-b, TIMP-1, TIMP-2, TNF-a, TNF-b, TPO, VEGF.Test Principle:This assay employs an antibody specific for human CEA coated on a 96-well plate. Standards and samples are pipetted into the wells and CEA present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-human CEA antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CEA bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450nm. The intra-Assay reproducibility is CV<10% & inter-Assay is CV<12%.Kit Components:Microplate (Item A) coated with anti-human CEA, 1x96 wells?Wash Buffer Concentrate (20x) (Item B), 1x25ml ?Standards (Item C) (recombinant human CEA), 2x vials ?Assay Diluent A (Item D) Standard/Sample (serum/plasma) in 0.09% sodium azide, 1x30ml ?Assay Diluent B (Item E) Standard/Sample (cell culture/urine) 5x concentrated, 1x15ml ?Detection Antibody CEA (Item F), biotinylated anti-human CEA, 2x vials HRP-Streptavidin Concentrate (Item G), 1000x concentrated, 1x200ul ?TMB One-Step Substrate Reagent (Item H) 3,3’,5,5’-tetramethylbenzidine in buffer, 1x12ml ?Stop Solution (Item I), 0.2M sulfuric acid, 1x8ml Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
