- This mouse CD40 BioAssay ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of mouse CD40. Sample Type:Serum, plasma and cell culture supernatantsIntended Use:For use in quantitative protein detection, establishing normal range and validation of antibody array results.Sensitivity:The minimum detectable dose of CD40 is typically <3pg/ml. Range:3-1500pg/mlSpecificity:Recognizes mouse CD40. This ELISA kit shows no cross-reactivity with the following cytokines tested: Mouse CD30, L CD30, T CD40, CRG-2, CTACK, CXCL16, Eotaxin , Eotaxin-2, Fractalkine, GCSF, GM-CFS, IFN-g, IGFBP-3, IGFBP-5, IGFBP-6, IL-1a, IL-1b, IL-2, IL-3, IL-3 Rb, IL-4, IL-5, IL-9, IL-10, IL-12 p40/p70, IL-12 p70, IL-13,IL-17, KC, Leptin R, LEPTIN(OB), LIX, L-Selectin, Lymphotactin, MCP-1, M, CP-5, MIG, MIP-1a, MIP-1g, MIP-2, MIP-3b, MIP-3a, PF-4, P-Selectin, RANTES, SCF, SDF-1a, TARC, TCA-3, TECK, TIMP-1, TNF-a, TNF RI, TNF RII, TPO, VCAM-1, VEGF.Test Principle:This assay employs an antibody specific for mouse CD40 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD40 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse CD40 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD40 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450nm. The intra-Assay reproducibility is CV<10% & inter-Assay is CV<12%.Kit Components:Microplate (Item A) coated with anti-mouse CD40, 1x96 wells?Wash Buffer Concentrate (20x) (Item B), 1x25ml?Standards (Item C) (recombinant mouse CD40), 2x vials?Assay Diluent A (Item D) for Standard/Sample (serum/plasma). 0.09% sodium azide, 1x30ml?Assay Diluent B (Item E) for Standard/Sample (cell culture medium). 5x concentrated, 1x15ml?Detection Antibody CD40 (Item F), biotinylated anti-mouse CD40, 2x vials?HRP-Streptavidin Concentrate (Item G), 500x concentrated, 1x200ul?TMB One-Step Substrate Reagent (Item H) 3,3’,5,5’-tetramethylbenzidine, in buffer, 1x12ml ?Stop Solution (Item I), 0.2M sulfuric acid, 1x8mlStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
