- The Ghrelin BioAssay Enzyme Immunoassay (EIA) Kit is an in vitro quantitative assay for detecting Ghrelin peptide based on the principle of Competitive Enzyme Immunoassay.Sample Type:Serum, plasma, cell culture media or other sample typesIntended Use:For use in quantitative protein detection, establishing normal range and validation of antibody array results.Sensitivity:The minimum detectable concentration of Ghrelin is <161pg/ml.Range:0.1-1000ng/mlSpecificity:Recognizes human, mouse and rat Ghrelin. This EIA kit shows no cross-reactivity with the following cytokines tested: e.g., Nesfatin, Angiotensin II, NPY and APC.Test Principle:The microplate in the BioAssay Enzyme Immunoassay (EIA) Kit is pre-coated with anti-rabbit secondary antibody. After a blocking step and incubation of the plate with anti-Ghrelin antibody, both biotinylated Ghrelin peptide and peptide standard or targeted peptide in samples interacts competitively with the Ghrelin antibody. Uncompeted (bound) biotinylated Ghrelin peptide then interacts with Streptavidin-horseradish peroxidase (SAHRP), which catalyzes a color development reaction. The intensity of colorimetric signal is directly proportional to the amount of biotinylated peptide-SA-HRP complex and inversely proportional to the amount of Ghrelin peptide in the standard or samples. This is due to the competitive binding of Ghrelin antibody between biotinylated Ghrelin peptide and peptides in standard or samples. A standard curve of known concentration of Ghrelin peptide can be established and the concentration of Ghrelin peptide in the samples can be calculated accordingly. The intra-Assay reproducibility is CV<10% & inter-Assay is CV<15%.Kit Components:Ghrelin Microplate (Item A) coated with secondary antibody, 1x96wells20X Wash Buffer Concentrate (Item B), 1x25mlLyophilized Standard Ghrelin Peptide (Item C), 2x vials Lyophilized anti-Ghrelin Polyclonal antibody (Item N), 2x vials 1X Assay Diluent E (Item R) for Standard/Sample, 2x25mlLyophilized Biotinylated Ghrelin peptide (Item F), 2x vialHRP-Streptavidin Concentrate (Item G), 100x concentrated, 1x600ul Lyophilized Positive Control (Item M) 1 vial TMB One-Step Substrate Reagent (Item H), 1x12mlStop Solution (Item I), 0.2M sulfuric acid, 1x8mlStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
