- The rat IL-2 BioAssay ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of rat IL-2. Sample Type:Serum and plasma (rat IL-2 conc. is pretty low in normal serum/plasma, it may not be detected in this assay). Cell culture supernatants.Intended Use:For use in quantitative protein detection, establishing normal range and validation of antibody array results.Sensitivity:The minimum detectable dose of IL-2 is typically <0.1ng/ml.Range:0.1-30ng/mlSpecificity:Recognizes rat IL-2. This ELISA kit shows no cross-reactivity with the following cytokines tested: rat CINC-2, CINC-3, CNTF, Fractalkine, IL-1a, IL-1b, IL-4, IL-6, IL-10, GM-CSF, IFN-g, Leptin, Lix, MCP-1, MIP-3a, b-NGF, TIMP-1, TNF-a.Test Principle:This BioAssay employs an antibody specific for IL-2 coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-2 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-rat IL-2 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-2 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450nm. The intra-Assay reproducibility is CV<10% & inter-Assay is CV<12%.Kit Components:IL-2 Coated Microplate (Item A), 1x96wells20X Wash Buffer Concentrate (Item B), 1x25ml Standards (Item C) (recombinant IL-2), 2x vials Assay Diluent A (Item D) for Standard/Sample (serum/plasma), 1x30ml Assay Diluent B (Item E) for Standard/Sample (cell culture medium), 5x conc., 1x15ml Detection Antibody IL-2 (Item F), biotinylated anti-IL-2, 2x vial HRP-Streptavidin Concentrate (Item G), 500x concentrated, 1x200ulTMB One-Step Substrate Reagent (Item H), 1x12ml Stop Solution (Item I), 0.2M sulfuric acid, 1x8mlStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
