- Sphingomyelinase (SMase) cleaves sphingomyelin to produce phosphocholine and ceramide. The activation of SMase leads to increased production of ceramide, which acts as a lipid second messenger that induces a variety of cell regulatory phenomena such as apoptosis, cell differentiation, cell proliferation, and sterol homeostasis. There are five distinct types of SMases.Sample Type:Animal tissues: brain, heart, kidney, etc. Cell culture: Adherent or suspension cells.Intended Use:For use in the measurement of SMase activity in various tissues/cells extracts and screening of SMase inhibitors.Sensitivity:This high-throughput adaptable assay kit can detect sphingomyelinase activity as low as 1.25mU/ml (~5 uU/mg tissue/cells) in a variety of samples.Test Principle:The SMase Activity BioAssay Kit provides a simple and sensitive method for measuring neutral-SMase enzymatic activity using colorimetry (OD 570nm). Neutral sphingomyelinase (N-SMase) is a Mg2+ sensitive enzyme that can be activated by a host of physiologically relevant and structurally diverse molecules. In this assay, SMase converts sphingomyelin substrate to phosphocholine and ceramide, which is then further utilized to produce an intermediate that reacts with a highly specific probe to generate color. Kit Components:SMase Assay Buffer, 1x28ml SMase Probe, DMSO, 1x200ul SMase Substrate, lyophilized, 1x vial SMase Enzyme Mix I, lyophilized, 1x vialSMase Enzyme Mix II, lyophilized, 1x vialCholine Standard, lyophilized, 1x vialSMase Positive Control, lyophilized, 1x vialSMase Extraction Detergent, 1x1mlStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
