- Chylomicron remnants and very low density lipoprotein (VLDL) remnants are rapidly removed from the circulation by receptor-mediated endocytosis in the liver. Apolipoprotein E, a main apoprotein of the chylomicron, binds to a specific receptor on liver cells and peripheral cells. ApoE is essential for the normal catabolism of triglyceride-rich lipoprotein constituents. The APOE gene is mapped to chromosome 19 in a cluster with APOC1 and APOC2. Defects in apolipoprotein E result in familial dysbetalipoproteinemia, or type III hyperlipoproteinemia (HLP III), in which increased plasma cholesterol and triglycerides are the consequence of impaired clearance of chylomicron and VLDL remnants.Sample Type:Serum, plasma and tissue homogenatesIntended Use:For the quantitative determination of porcine apolipoprotein E (Apo-E) concentrations in serum, plasma and tissue homogenates. Sensitivity:7.81ng/mlRange:31.25-2000ng/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of porcine Apo-E. No significant cross-reactivity or interference between porcine Apo-E and analogues was observed.Test Principle:This BioAssay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with Apo-E. Standards or samples are added to the appropriate microtiter plate wells with Horseradish Peroxidase (HRP) conjugated antibody preparation specific for Apo-E. The competitive inhibition reaction is launched between with pre-coated Apo-E and Apo-E in samples. A substrate solution is added to the wells and the color develops in opposite to the amount of Apo-E in the samples. The color development is stopped and the intensity of the color is measured.
