- Biotin, BioAssay ELISA Kit is a competitive ELISA intended for the quantitative determination of Biotin in human and mouse serum and plasma.Detection Range:12.5-200pg/mlSensitivity:<4.986pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest Biotin concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.Intra-assay Precision: %CV <8 Inter-assay Precision: %CV <10Specificity:This assay has high sensitivity and excellent specificity for detection of Biotin. No significant cross-reactivity or interference between biotin and analogues was observed.Test Principle:This BioAssay employs the direct competitive inhibition enzyme immunoassay technique. The microplate was pre-coated with Avidin. Add Biotin (Standards or samples) to the well, and then add HRP-conjugated Biotin. A competitive inhibition reaction is launched between Biotin (Standards or samples) and HRP conjugated Biotin with the Avidin. The substrate solutions are added to the wells, respectively. And the color develops in opposite to the amount of Biotin bound in the initial step. The color development is stopped and the intensity of the color is measured. Kit Components:227542A: Microtiter Strips, 1x48 wells or 1x96 wells*227542B: Standard, 200pg/ml, 1x100ul or 1x200ul*227542C: HRP Conjugate, 100X, 1x30ul or 1x60ul227542D: HRP Diluent, 1x10ml or 1x20ml227542E: Sample Diluent, 1x20ml or 1x40ml227542F: Wash Buffer, 25X, 1x20ml227542G: TMB Substrate, 1x10ml or 1x20ml227542H: Stop Solution, 1x10ml or 1x20mlNote: The Stop Solution (227542H) provided with this kit is an acid solution. Wear eye, hand, face, and clothing protection when using this material.Storage and Stability:Store *227542B and *227542C at -20oC. Store all other kit components at 4°C. Stable for 6 months after receipt. Once opened, the kit is stable for 1 month providing this is within the expiration date of the kit. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.Assay Summary:1. Prepare reagents and samples as instructed.2. Set a Blank well without any solution3. Add 50ul Standard or sample to each well.4. Add 50ul HRP Conjugate, 1X to each well (Not to the Blank well).5. Incubate for 40 minutes at 37°C.6. Aspirate and wash 5 times.7. Add 90ul TMB Substrate to each well.8. Incubate for 20 minutes at 37°C. Protect from light.9. Add 50ul Stop Solution to each well. Read at 450nm within 10 minutes.
