- Folic Acid (FA), BioAssay ELISA Kit is a competitive ELISA intended for the quantitative determination of FA concentration in human serum and plasma.Detection Range:12.5-200pg/ml Sensitivity:<6.25pg/mlPrecision:Intra-assay Precision: CV: <8%Inter-assay Precision: CV: <10%Principle of the Assay:This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to FA. Standards or samples and FA-HRP conjugate are added to the appropriate microtiter plate wells and a competitive inhibition reaction is launched between with HRP-conjugated FA and FA in the standards or samples. A substrate solution is added to the wells. The color development is stopped and the intensity of the color is measured. The intensity of the color is inversely proportional to the amount of FA in the samples.Kit Components:227665A: Microtiter Strips, 1x96 wells. Pre-coated; ready to use. 227665B: Standard (10X concentrate), 1x200ul227665C: HRP Conjugate (100X Concentrate), 1x60ul227665D: HRP Conjugate Diluent, 1x10ml227665E: Sample Diluent 2x20ml227665F: Wash Buffer (25X Concentrate), 1x20ml227665G: TMB Substrate, 1x10ml227665H: Stop Solution, 1x10ml227665J: Wash Buffer (25X Concentrate), 1x20mlStorage and Stability:Store all kit components at 4°C. Unused kit is stable for 6 months after receipt. It is recommended to store the coated plate and standard at -20°C if the kit will not be used within one month. Once opened, the kit is stable for 1 month providing this is within the expiration date of the kit. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.Sample Preparation and Storage:Serum: Use a serum separator tube (SST) and allow samples to clot for two hours at RT or overnight at 4°C before centrifugation for 15 minutes at 1000 ×g. Remove serum and assay immediately or aliquot and store samples at -20°C or -80°C. Avoid repeated freeze-thaw cycles.Plasma: Collect plasma using EDTA or heparin as an anticoagulant. Centrifuge for 15 minutes at 1000×g at 4°C within 30 minutes of collection. Assay immediately or aliquot and store samples at -20°C or -80°C. Avoid repeated freeze-thaw cycles. We recommend that serum or plasma samples be diluted with Sample Diluent before the test. The suggested 100-fold dilution can be achieved by adding 10ul sample to 40ul of Sample Diluent. Complete the 100-fold dilution by adding 15ul of this solution to 285ul of Sample Diluent. The recommended dilution factor is for reference only. The optimal dilution factor should be determined by users according to their particular experiments.Assay Summary:1. Prepare reagents and samples as instructed.2. Set a Blank well without any solution3. Add 50ul Standard or sample to each well.4. Add 50ul HRP Conjugate to each well (do not add any to the blank well).5. Incubate for 40 minutes at 37°C.6. Aspirate and wash 5 times.7. Add 90ul TMB substrate to each well8. Incubate for 20 minutes at 37°C. Protect from light12. Add 50ul Stop Solution to each well. Read at 450nm within 5 minutes.
