- Sample Type:Serum and plasmaIntended Use:For the quantitative determination of endogenic rat free thyroxine (FT4) concentrations in serum and plasma. Sensitivity:1.08pmol/LRange:4-64pmol/LSpecificity:This assay has high sensitivity and excellent specificity for detection of rat FT4. No significant cross-reactivity or interference between rat FT4 and analogues was observed.Test Principle:This BioAssay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to T4. Standards or samples are added to the appropriate microtiter plate wells with Biotin-conjugated T4. A competitive inhibition reaction is launched between T4 (Standards or samples) and Biotin-conjugated T4 with the pre-coated antibody specific for T4. The more T4 in samples, the less antibody bound by Biotin-conjugated T4. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Substrate solution is added to the wells and the color develops in opposite to the amount of FT4 in the sample. The color development is stopped and the intensity of the color is measured. Kit Components:Assay plate, 1x96 wellsStandard, 5x1ml Conjugate, 1x6ml HRP-avidin, 1x6ml Wash Buffer (20X), 1x15ml Substrate A, 1x7ml Substrate B, 1x7ml Stop Solution, 1x7ml Adhesive Strip (For 96 wells), 4x strips Instruction manual Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
