- This protein is a member of the somatotropin/prolactin family of hormones which play an important role in growth control. The gene, along with four other related genes, is located at the growth hormone locus on chromosome 17 where they are interspersed in the same transcriptional orientation; an arrangement which is thought to have evolved by a series of gene duplications. The five genes share a remarkably high degree of sequence identity. Alternative splicing generates additional isoforms of each of the five growth hormones, leading to further diversity and potential for specialization. This particular family member is expressed in the pituitary but not in placental tissue as is the case for the other four genes in the growth hormone locus. Mutations in or deletions of the gene lead to growth hormone deficiency and short stature.Sample Type:Serum and plasmaIntended Use:For the quantitative determination of human growth hormone (GH) concentrations in serum and plasma. Sensitivity:1ng/mlRange:2.5-50ng/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of human GH. No significant cross-reactivity or interference between human GH and analogues was observed.Test Principle:This BioAssay employs the quantitative sandwich enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with biotin-BSA and Avidin. Standards and samples are pipetted into the wells with a biotin-conjugated antibody specific for GH. After washing, GH antibody conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GH in sample. The color development is stopped and the intensity of the color is measured. Kit Components:Assay plate, 1x96 wellsStandard, 6x1ml Antibody, 1x6ml HRP-conjugate, 1x10ml Wash Buffer (10X), 1x20ml Substrate A, 1x7ml Substrate B, 1x7ml Stop Solution, 1x7ml Adhesive Strip (For 96 wells), 4x strips Instruction manual Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
