- After removal of the precursor signal peptide, proinsulin is post-translationally cleaved into two chains (peptide A and peptide B) that are covalently linked via two disulfide bonds. Binding of this mature form of insulin to the insulin receptor (INSR) stimulates glucose uptake. A variety of mutant alleles with changes in the coding region have been identified. There is a read-through gene, INS-IGF2, which overlaps with this gene at the 5 region and with the IGF2 gene at the 3 region.Sample Type:Serum and plasmaIntended Use:For the quantitative determination of sheep insulin (INS) concentrations in serum and plasma. Sensitivity:5uIU/mlRange:8-140uIU/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of sheep INS. No significant cross-reactivity or interference between sheep INS and analogues was observed.Test Principle:This BioAssay employs the quantitative sandwich enzyme immunoassay technique. An antibody specific for INS has been pre-coated onto a microplate. Standards and samples are pipetted into the wells with a Horseradish Peroxidase (HRP) conjugated antibody specific for INS. Following a wash to remove any unbound reagent, a substrate solution is added to the wells and color develops in proportion to the amount of INS bound in the initial step. The color development is stopped and the intensity of the color is measured. Kit Components:Assay plate, 1x96 wellsStandard, 6x0.5ml HRP-conjugate, 1x6ml Wash Buffer (20X), 1x15ml Substrate A, 1x7ml Substrate B, 1x7ml Stop Solution, 1x7ml Adhesive Strip (For 96 wells), 4x strips Instruction manual Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
