產品描述: This gene encodes a protein that is secreted by white adipocytes, and which plays a major role in the regulation of body weight. This protein, which acts through the leptin receptor, functions as part of a signaling pathway that can inhibit food intake and/or regulate energy expenditure to maintain constancy of the adipose mass. This protein also has several endocrine functions, and is involved in the regulation of immune and inflammatory responses, hematopoiesis, angiogenesis and wound healing. Mutations in this gene and/or its regulatory regions cause severe obesity, and morbid obesity with hypogonadism. This gene has also been linked to type 2 diabetes mellitus development.Sample Type:Serum, plasma and tissue homogenatesIntended Use:For the quantitative determination of human leptin (LEP) concentrations in serum, plasma and tissue homogenates. Sensitivity:0.060ng/mlRange:0.156-10ng/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of human LEP. No significant cross-reactivity or interference between human LEP and analogues was observed.Test Principle:This BioAssay employs the quantitative sandwich enzyme immunoassay technique. An antibody specific for LEP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any LEP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for LEP is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of LEP bound in the initial step. The color development is stopped and the intensity of the color is measured. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
