- Tri-Iodothyronine (T3), BioAssay ELISA Kit is a competitive ELISA intended for the quantitative determination of T3 in fish serum, plasma and tissue homogenates.Detection Range:0.5-8ng/mlIntra-assay Precision: %CV <15 Inter-assay Precision: %CV <15Sensitivity:The minimum detectable dose of fish T3 is typically less than 0.5ng/ml.The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest fish T3 concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.Specificity:This assay has high sensitivity and excellent specificity for detection of figh T3. No significant crossreactivity or interference between fish T3 and analogs was observed.Note: Limited by current skills and knowledge, it is impossible for us to complete the crossreactivity detection between fish T3 and all the analogs. Therefore, crossreaction may still exist.Kit Components:227997A: Microtiter Strips, 1x48 wells or 1x96 wells227997B: Standard, 0.5ng/ml, 1x500ul or 1x1ml227997C: Standard, 1ng/ml, 1x500ul or1x1ml227997D: Standard, 2ng/ml, 1x500ul or1x1ml227997E: Standard, 4ng/ml, 1x500ul or 1x1ml227997F: Standard, 8ng/ml, 1x500ul or1x1ml227997G: T3 (Biotin), 1x3ml or 1x6ml227997H: Avidin (HRP), 1x3ml or 1x6ml227997J: Wash Buffer, 20X, 1x15ml or 1x30ml 227997K: Substrate A, 1x3.5ml or 1x7ml227997L: Substrate B, 1x3.5ml or 1x7ml227997M: Stop Solution, 1x3.5ml or 1x7mlNote: The Stop Solution (227997M) provided with this kit is an acid solution. Wear eye, hand, face, and clothing protection when using this material.Storage and Stability:Store all kit components at 4°C. Stable for 6 months after receipt. Once opened, the kit is stable for 1 month providing this is within the expiration date of the kit. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.Assay Summary:1. Prepare reagents and samples as instructed.2. Set a Blank well without any solution3. Add 50ul Standard or sample to each well.4. Add 50ul T3 (Biotin) to each well (Not to the Blank well).5. Incubate for 60 minutes at 37°C.6. Aspirate and wash 3 times.7. Add 50ul Avidin (HRP) to each well (Not to the Blank well).8. Incubate for 30 minutes at 37°C. 9. Aspirate and wash 3 times,10. Add 50ul Substrate A and 50ul Substrate B to each well. 11. Incubate for 15 minutes at 37°C. Protect from light12. Add 50ul Stop Solution to each well. Read at 450nm within 10 minutes.
