- EHMT1 (GLP) is a histone methyltransferase that exhibits methylation activity toward H3-K9.Intended Use:The EHMT1 (GLP) Direct Activity Chemiluminescent BioAssay Kit is designed to measure EHMT1, also known as G9a-like protein or GLP activity for screening and profiling applications. Test Principle:The key to the EHMT1 (GLP) Chemiluminescent BioAssay Kit is a highly specific antibody that recognizes methylated K9 residue of H3. With this kit, only three simple steps on a microtiter plate are required for methyltransferase detection. First, S-adenosylmethionine is incubated with a sample containing assay buffer and methyltransferase enzyme for one hour. Next, primary antibody is added. Finally, the plate is treated with an HRP-labeled secondary antibody followed by addition of the HRP substrate to produce chemiluminescence that can then be measured using a chemiluminescence reader.Contraindications:DMSO >1%, strong acids or bases, ionic detergents, high saltApplications:Great for studying enzyme kinetics, screening small molecular inhibitors for drug discovery and HTS applications.Kit Components:Plate precoated with histone substrate, 1x96 wellsEHMT1 (GLP), 1x4ug Primary antibody 5, 1x12.5ul Secondary HRP-labeled antibody 1, 1x10ul 20uM S-adenosylmethionine, 1x250ul 4x HMT assay buffer 1, 1x3ml Blocking buffer, 1x50ml HRP chemiluminescent substrate, 2 components x 6ml each Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
