產品描述: The FBXL10 Chemiluminescent BioAssay Kit is designed to measure FBXL10 activity for screening and profiling applications. Test Principle:FBXL10, also known as KDM2B, JHDM1B, and CXXC2, is a histone lysine demethylase that exhibits demethylation activity toward trimethyllysine 4 (H3K4me3) and dimethyl-lysine 36 (H3K36me2) on histone H3. The key to the FBXL10 Chemiluminescent BioAssay Kit is a highly specific antibody that recognizes demethylated substrate. With this kit, only three simple steps on a microtiter plate are required for detection of demethylase activity. First, FBXL10 enzyme is incubated with the methylated H3 peptide for one hour. Next, primary antibody is added. Finally, the plate is treated with an HRP-labeled secondary antibody followed by the addition of the HRP substrate to produce chemiluminescence that can be measured using a chemiluminescence reader.Contraindications:DMSO >1%, strong acids or bases, ionic detergents, high saltApplications:Great for studying enzyme kinetics and HTS applications.Kit Components:8-well strip plate module precoated with histone substrate, 1x plateFBXL10, 1x20ugPrimary antibody 16, 1x12.5ul Secondary HRP-labeled antibody 2, 1x10ul4x FBXL10 assay buffer, 1x3ml Blocking buffer, 1x50ml HRP chemiluminescent substrate A, (transparent bottle), 1x6ml HRP chemiluminescent substrate B, (brown bottle), 1x6ml Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
