- Intended Use:The MLL1 Complex Direct Activity Chemiluminescent BioAssay kit is designed to measure MLL1 activity for screening and profiling applications, using purified MLL1 and its complex components: WDR5, RbBP5, Ash2, and DPY30. Test Principle:The key to the MLL1 Complex Chemiluminescent BioAssay Kit is a highly specific antibody that recognizes methylated K4 residue of Histone H3. With this kit, only three simple steps on a microtiter plate are required for methyltransferase detection. First, S-adenosylmethionine is incubated with a sample containing assay buffer and methyltransferase enzyme for one hour. Next, primary antibody is added. Finally, the plates are treated with an HRP-labeled secondary antibody followed by the addition of the HRP substrate to produce chemiluminescence that can be measured using a chemiluminescence reader.Contraindications:DMSO >1%, strong acids or bases, ionic detergents, high saltApplications:Great for studying enzyme kinetics, screening small molecular inhibitors for drug discovery and HTS applications.Kit Components:Plate, 1x96 wellsMLL1/WDR5/Ash2L/RbBP5/DPY30, 1x25ug 20uM S-adenosylmethionine, 1x250ul Primary antibody 2, 1x12.5ul 4x HMT assay buffer 1, 1x3ml Secondary HRP-labeled antibody 1, 1x10ul Blocking buffer, 1x50ml HRP chemiluminescent substrate, 2 components x 6ml each Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
