- Intended Use:The NSD2 Direct Activity Chemiluminescent BioAssay Kit is designed to measure NSD2 activity for screening and profiling applications.Test Principle:The key to the NSD2 Chemiluminescent Activity BioAssay Kit is a highly specific antibody that recognizes methylated residue of Histone H3. With this kit, only three simple steps are required for methyltransferase detection. First, S- adenosylmethionine is incubated with a sample containing assay buffer and methyltransferase enzyme. Next, primary antibody is added. Finally, the plate is treated with an HRP-labeled secondary antibody followed by addition of the HRP substrate to produce chemiluminescence that can then be measured using a chemiluminescence reader.Contraindications:DMSO >1%, strong acids or bases, ionic detergents, high saltApplications:Great for studying enzyme kinetics, screening small molecular inhibitors for drug discovery and HTS applications.Kit Components:Plate precoated with histone substrate, 1x96 wellsNSD2 human enzyme, 1x50ug S-adenosylmethionine, 100uM, 1x250ul Primary antibody 16, 1x12.5ul Secondary HRP-labeled antibody 2, 1x10ul HMT assay buffer 7, 4x, 1x3ml Blocking buffer, 1x50ml HRP chemiluminescent substrate A, (translucent bottle), 1x6ml HRP chemiluminescent substrate B, (brown bottle), 1x6ml Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
