- PARP2 is known to catalyze the NAD-dependent addition of poly (ADP-ribose) to histones.Intended Use:The PARP2 Chemiluminescent Activity BioAssay kit is designed to measure PARP2 activity for screening and profiling applications. Test Principle:The key to the PARP2 Chemiluminescent Activity BioAssay is the biotinylated substrate. With this kit, only three simple steps are required for PARP2 reactions. First, histone proteins are coated on a 96-well plate. Next, the PARP2 biotinylated substrate is incubated with the assay buffer that contains the PARP2 enzyme. Finally, the plate is treated with streptavidin-HRP followed by addition of the HRP substrate to produce a chemiluminescence that can then be measured using a chemiluminescence reader. This kit is also suitable for use with cell extracts, but nonspecific ribosylation activity may be detected.Contraindications:DMSO >1%, strong acids or bases, ionic detergents, high saltApplications:Great for studying enzyme kinetics, screening small molecular inhibitors for drug discovery and HTS applications.Kit Components:Module plate, 1x96 wellsPARP2, 1x5ug Histone mixture, 5x, 1x1ml Assay Mixture Containing Biotinylated Substrate, 10x, 1x300ulPARP assay buffer, 10x, 1x1ml Blocking buffer, 1x25ml Activated DNA (5x), 1x500ul Streptavidin-HRP, 1x100ul HRP chemiluminescent substrate A, (translucent bottle), 1x6ml HRP chemiluminescent substrate B, (brown bottle), 1x6ml Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
