- PRMT4 (CARM1) is a histone methyltransferase that exhibits methylation activity toward H3-R17.Intended Use:The PRMT4 Direct Activity Chemiluminescent BioAssay kit is designed to measure PRMT4/CARM1 activity activity for screening and profiling applications. Test Principle:The key to the PRMT4 Chemiluminescent BioAssay Kit is a highly specific antibody that recognizes methylated PRMT4 substrate. With this kit, only three simple steps on a microtiter plate are required for methyltransferase detection. First, S-adenosylmethionine is incubated with a sample containing assay buffer and methyltransferase enzyme. Next, primary antibody is added. Finally, the strip plates are treated with an HRP-labeled secondary antibody followed by addition of an HRP substrate to produce chemiluminescence that can then be measured using a chemiluminescence reader.Contraindications:DMSO >1%, strong acids or bases, ionic detergents, high saltApplications:Great for studying enzyme kinetics, screening small molecular inhibitors for drug discovery and HTS applications.Kit Components:Plate precoated with PRMT4 substrate, 1x96 wellsPRMT4/CARM1 enzyme, 1x20ug S-adenosylmethionine, 20um, 1x250ul Primary antibody 9, 1x100ul Secondary HRP-labeled antibody 2, 1x10ul 1% BSA, 1x50ul HMT assay buffer 5, 4x, 1x3ml Blocking buffer, 1x50ml HRP chemiluminescent substrate, 2 components x 6ml each Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
