產品描述: Histone Acetyltransferases (HATs) are enzymes that acetylate histone substrates resulting in important regulatory effects on chromatin structure and assembly, and gene transcription. Modifications of these proteins by HATs play an important role in the control of gene expression, and their dysregulation has been linked to cancer, neurodegeneration, and other diseases.Sample Type:Nuclear extracts from cells and tissue, recombinant enzymes.Intended Use:For measurement of HAT activity in nuclear extracts and purified enzyme preparations.Sensitivity:6mUTest Principle:This HAT Activity BioAssay Kit utilizes Acetyl CoA and H3 histone peptide as substrates. In this assay, HAT enzyme catalyzes the transfer of acetyl groups from Acetyl-CoA to the histone peptide, thereby generating two products - acetylated peptide and CoA-SH. The CoA-SH reacts with the developer to generate a product that is detected fluorometrically at Ex/Em= 535/587nm.Kit Components:HAT Assay Buffer, 1x25ml Acetyl CoA, Lyophilized, 1x vial H3 Peptide, Lyophilized, 1x vialSubstrate Mix, Lyophilized, 1x vial Developer, 1x100ulPicoProbe, 1x200ulCoA Standard, Lyophilized, 1x vialPositive Control, HeLa Nuclear Extract, 1x40ulStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
