- Human Osteopontin (OPN) ELISA BioAssay Kit is an in vitro assay for the quantitative measurement of human OPN. OPN is a multifunctional, hyperphosphorylated acidic glycoprotein. As a key component of extracellular matrix-associated protein abundantly expressed in bone, OPN is a key regulator of bone mineralization. In addition, OPN is expressed in multiple other cell types including macrophages, smooth muscle cells and hepatocytes. OPN up-regulation occurs upon exposure to pro-inflammatory cytokines (TNF-alpha, TGF-beta etc.) or under pathological conditions such as hypoxia and hyperglycemia. OPN also functions as a chemokine-like protein involved in regulating cell proliferation, inflammatory responses, angiogenesis and metastasis, and is critical in the development of multiple disorders including cardiovascular disease, diabetes, and cancer. Sample Type:Plasma, serum, and cell culture supernatantsIntended Use:Quantitative measurement of native and recombinant human Osteopontin (OPN).Sensitivity:0.2ng/mlRange:0.2-70ng/ml Specificity:Native and recombinant human OPN. This ELISA kit shows no cross-reactivity with other relevant proteins.Test Principle:The BioAssay employs monoclonal antibody specific for human OPN coated on a 96-well microplate. Standards and samples are pipetted into the wells and OPN present in the sample is bound to the wells by the immobilized antibody. The antibody-captured OPN is then detected by a biotinylated monoclonal antibody. With HRP conjugated Streptavidin and a HRP substrate, amount of OPN in the sample is determined by measuring absorbance of the HRP product at 450nm with a microplate reader. The intra-assay reproducibility as measured by the coefficient of variation (CV) is <5% and inter-assay CV <7%.Kit Components:Plate coated with OPN MAb, 1x96 wellsAssay Diluent A, 1x20ml Assay Diluent B, 1x20ml Wash Buffer/Ab Diluent, 10X, 1x20mlrh OPN Standard, 500ng/vial, 3x2ul Detection Ab, 100X, 1x100ul HRP Conjugate, 100X, 1x140ul TMB Substrate, 1x11ml Stop Solution, 1x11ml Plate Sealer, 2x sealers Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
