- Renin (EC 3.4.23.15), also known as angiotensinogenase, is an enzyme that participates in the renin-angiotensin system (RAS) which mediates extracellular volume (i.e. blood plasma, lymph and interstitial fluid), and arterial vasoconstriction. In addition to the systemic RAS, locally expressed RAS has been found in the kidney, heart and nervous system. An overactive renin-angiotensin system leads to vasoconstriction and retention of sodium and water, causing hypertension. Renin inhibitors are widely used for the treatment of hypertension. Sample Type:Crude extracts, purified recombinant protein and rissue lysates.Intended Use:For use in the detection of rat Renin activity Sensitivity:0.12mu or ~0.1ug/ml Test Principle:In this Rat Renin Activity BioAssay Kit, rat Renin and other proteases hydrolyze a FRET substrate resulting in fluorescence at Ex/Em=328/552nm. In the presence of a rat Renin-specific inhibitor, hydrolysis of the substrate is due to only the activity of other proteases. The difference between the total activity and the activity in the presence of rat Renin-specific inhibitor gives the rat Renin activity in the sample. Kit Components:Renin Assay Buffer, 1x25ml ?Homogenization Buffer, 1x60ml ?Rat Renin Substrate, 1x200ul?Rat Renin, lyophilized, 1x vial?Rat Renin Inhibitor, lyophilized, 1x vial?EDANS Standard, 100um, 1x100ulStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
