- Interferon -gamma (IFN-gamma) exerts a wide range of immunoregulatory activities and is considered to be the prototype proinflammatory cytokine. Mature mouse IFNgamma exists as a noncovalently linked homodimer of 20-25kD variably glycosylated subunits. It shares 86% amino acid sequence identity with rat IFN-gamma and 38%--44% with human, porcine, and rhesus IFN-gamma. IFNgamma is produced by a variety of immune cells under inflammatory conditions, notably by T cells and NK cells. It plays a key role in host defense by promoting the development and activation of Th1 cells, chemoattraction and activation of monocytes and macrophages, up-regulation of antigen presentation molecules, and immunoglobulin class switching in B cells. It also exhibits antiviral, antiproliferative, and apoptotic effects.Sample Type:Serum and plasmaIntended Use:The kit is a sandwich enzyme immunoassay for the in vitro quantitative measurement of IFN-gamma in mouse serum, plasma; and other biological fluids not being tested.Sensitivity:4pg/mlRange:16.4-4000pg/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of mouse IFN-gamma.Precision:Intra CV<10%; Inter CV <15%Test Principle:This assay employs the fast quantitative sandwich enzyme immunoassay technique. Antibody specific for mouse IFN-gamma has been pre-coated onto a microplate. Standards, samples and mouse IFN-gamma mix are pipetted into the wells and any IFN-gamma present is bound by the immobilized antibody. Following a wash to remove any unbound reagent, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IFN-gamma bound in the initial step.
