- IL-1a and IL-1b are structurally related polypeptides that share approximately 17% amino acid (aa) identity in mouse. Both proteins are produced by a wide variety of cells in response to inflammatory agents, infections, or microbial endotoxins. While IL-1a and IL-1b are regulated independently, they bind to the same receptor and exert identical biological effects. IL-1 RII has high affinity for IL-1b but functions as a decoy receptor and negative regulator of IL-1b activity. The mouse IL-1b cDNA encodes a 269aa precursor. A 117aa propeptide is cleaved intracellularly by the cysteine protease IL-1b -converting enzyme (Caspase-1/ICE) to generate the active cytokine. The 17kD mature mouse IL-1b shares 90% aa sequence identity with cotton rat and rat and 65%--78% identity with human. Sample Type:Serum and plasmaIntended Use:The kit is a sandwich enzyme immunoassay for the in vitro quantitative measurement of IL-1 beta in mouse serum, plasma; and other biological fluids not being tested.Sensitivity:4pg/mlRange:81.9-20,000pg/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of mouse IL-1 beta.Precision:Intra CV<10%; Inter CV <15%Test Principle:This assay employs the fast quantitative sandwich enzyme immunoassay technique. Antibody specific for mouse IL-1 beta has been pre-coated onto a microplate. Standards, samples and mouse IL-1 beta mix are pipetted into the wells and any IL-1 beta present is bound by the immobilized antibody. Following a wash to remove any unbound reagent, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-1 beta bound in the initial step.
