- IL-1ra was originally isolated from the urine of patients with monocytic leukemia and has also been purified from adherent monocytes. The protein shows 26% amino acid homology to IL-1b and 19% homology to IL-1a. It will compete with either factor for receptor binding, but does not interact with either one. Human IL-1ra will bind to both types of IL-1 receptor (I and II) on human cells. In mouse, IL-1 RII does not bind IL-1ra. IL-1ra has been shown to block the inflammatory responses induced by IL-1 both in vitro and in vivo. Pre-clinical and clinical studies were done to test possible therapeutic applications for IL-1ra in the treatment of sepsis, rheumatoid arthritis and chronic myelogenous leukemia.Sample Type:Serum and plasmaIntended Use:The kit is a sandwich enzyme immunoassay for the in vitro quantitative measurement of lL-1ra in mouse serum, plasma; and other biological fluids not being tested.Sensitivity:<1pg/mlRange:98.3-24,000pg/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of mouse IL-1ra.Precision:Intra CV<10%; Inter CV <15%Test Principle:This assay employs the fast quantitative sandwich enzyme immunoassay technique. Antibody specific for mouse IL-1ra has been pre-coated onto a microplate. Standards, samples and mouse IL-1ra mix are pipetted into the wells and any IL-1ra present is bound by the immobilized antibody. Following a wash to remove any unbound reagent, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-1ra bound in the initial step.
