- Tumor necrosis factor alpha (TNF-a) is the prototypic ligand of the TNF superfamily. It is a pleiotropic molecule that plays a central role in inflammation, immune system development, apoptosis, and lipid metabolism. Human TNF-a is synthesized as a 26kD type II transmembrane protein. It is produced by a wide variety of immune, epithelial, endothelial, and tumor cells. Sample Type:Serum and plasmaIntended Use:The kit is a sandwich enzyme immunoassay for the in vitro quantitative measurement of TNF-alpha in human serum, plasma; and other biological fluids not being tested.Sensitivity:<1pg/mlRange:187.5-12,000pg/mlSpecificity:This assay has high sensitivity and excellent specificity for detection of human TNF-alpha.Precision:Intra CV<10%; Inter CV <15%Test Principle:This assay employs the fast quantitative sandwich enzyme immunoassay technique. Antibody specific for TNF-alpha has been pre-coated onto a microplate. Standards, samples and human TNF-alpha mix are pipetted into the wells and any TNF-alpha present is bound by the immobilized antibody. Following a wash to remove any unbound reagent, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TNF-alpha bound in the initial step. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
