- Plasminogen (PLG), BioAssay ELISA Kit (Mouse) is a competitive ELISA intended for the quantitative determination of PLG concentration in mouse serum, plasma and tissue homogenates.Sensitivity:<15.6ng/mlRange:62.5-4000ng/mlPrecision:Intra-assay CV: <8%Inter-assay CV: <10%Principle of the Assay:This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with PLG. Standards or samples are added to the appropriate microtiter plate wells with HRP-conjugated anti-PLG antibody. A competitive inhibition reaction is launched between pre-coated PLG and PLG in the standards or samples. A substrate solution is added to the wells; the color development is stopped and the intensity of the color is measured. The intensity of the color is inversely proportional to the amount of PLG in the samples.Kit Components:227665A: Microtiter Strips (12 x 8 microwells). Pre-coated with PLG; ready to use. 227665B: Standard (Lyophilized), 2x1 vials227665C: HRP Conjugate (100X Concentrate), 1x60ul227665D: HRP Conjugate Diluent, 1x10ml227665E: Sample Diluent 2x20ml227665F: Wash Buffer (25X Concentrate), 1x20ml227665G: TMB Substrate, 1x10ml227665H: Stop Solution, 1x10ml227665J: Wash Buffer (25X Concentrate), 1x20mlStorage and Stability:Unopened kit: Store at 4°C. Unused kit is stable for 6 months after receipt. Opened Kit: Once opened, the following kit components are stable for up to 1 month at 4°C providing this is within the expiration date of the kit: HRP conjugate diluent, Sample diluent, Wash buffer, TMB substrate and Stop solution. The coated plate, standard and HRP conjugate may be stored at 4°C for up to one month; store the standard and HRP conjugate at -20°C if the kit will not be used within one month. For maximum recovery of product, centrifuge the original vials after thawing and prior to removing the cap.Assay Summary:1. Prepare reagents, working standards and samples as instructed.2. Set a Blank well without any solution3. Add 50ul Standard or sample to each well.4. Add 50ul HRP Conjugate to each well (do not add any to the blank well).5. Incubate for 60 minutes at 37°C.6. Aspirate and wash 5 times.7. Add 90ul TMB substrate to each well8. Incubate for 20 minutes at 37°C. Protect from light12. Add 50ul Stop Solution to each well. Read at 450nm within 5 minutes.
