- Interleukin 1 receptor, type II (IL1R2) also known as CD121b (Cluster of Differentiation 121b) is an interleukin receptor. The protein encoded by this gene is a cytokine receptor that belongs to the interleukin-1 receptor family. It is mapped to 2q11.2. IL1R2 binds interleukin alpha(IL1A), interleukin beta (IL1B), and interleukin 1 receptor, type I (IL1R1/IL1RA), and acts as a decoy receptor that inhibits the activity of its ligands. IL1R2 also modulates cellular response through non-signaling association with IL1RAP after binding to IL1B. The secreted IL1R2 recruits secreted IL1RAP with high affinity, and this complex formation may be the dominant mechanism for neutralization of IL1B by secreted/soluble receptors.Sample Type:Cell culture supernates and serum.Intended Use:For quantitative detection of human IL1R2 in cell culture supernates and serum.Sensitivity:<10pg/mlRange:31.2-2000pg/mlSpecificity:Natural and recombinant human IL1R2Test Principle:This human IL1R2 BioAssay ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for IL1R2 has been precoated onto 96-well plates. Standards(sf21) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for IL1R2 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human IL1R2 amount of sample captured in plate.Kit Components:96-well plate precoated with human IL1R2 antibody, 1Lyophilized recombinant human IL1R2 standard, 10ng/tubex2Biotinylated human IL1R2 antibody, 130ul (dilution 1:100)Avidin-Biotin-Peroxidase Complex(ABC), 130ul (dilution 1:100)Sample diluent buffer, 30mlAntibody diluent buffer, 12mlABC diluent buffer, 12mlTMB color developing agent, 10mlTMB stop solution, 10mlStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
