Anti-T-cell surface glycoprotein CD4 antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-T-cell surface glycoprotein CD4 antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the T-cell surface glycoprotein CD4 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of T-cell surface glycoprotein CD4 can be calculated.Kit Components:Microtiter Strips, 1x96 wells (8x12 wells)Standard, 2x1 vialSample/Standard Dilution Buffer, 1x20mlAntibody (Biotin) (Concentrated), 1x120ulAntibody Dilution Buffer, 1x10mlStreptavidin (HRP) (SABC), 1x120ulSABC Dilution Buffer, 1x10mlTMB Substrate, 1x10mlStop Solution, 1x10mlWash Buffer, 25X, 1x30mlStorage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for at least 6 months For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
