- Valsartan (Diovan, Tareg, Provas, Kalpress) is a synthetic antagonist of the Angiotensin II type 1 receptor. It has an effective concentration of 1-100 nM and an IC50 of 1.4 nM. Valsartan is administered orally for the treatment of mild to moderate essential hypertension. It causes a dose-dependent decrease in blood pressure. It is rapidly absorbed and reaches peak concentration 1-2 hours after intake. It has a half-life of 7-8 hours and is mostly excreted from the body in the stool. The recommended starting dose is 80 mg and no dose regulation is required for patients with creatinine clearance larger than 10 ml/min. In rat myocardial infarction (MI) models, Valsartan has been found to reduce left ventricle end-diastolic pressure suggesting it has a beneficial effect on myocardium dilatation following MI. Valsartan’s adverse effects include headache, fatigue, dizziness, abdominal pain and nausea with no age, gender or origin differences. More rare adverse effects include elevated serum potassium levels, neutropenia and anemia
